A New Coenzyme Q10 Preparation

Coenzyme Q10 is commercially available as a nutritional supplement
in various dosage forms. None of the dosage forms tested to date are
able to provide consistent and adequate plasma CoQ10 values. In addition,
because of the extremely poor solubility of CoQ10 in aque ous systems,
the currently available CoQ10 products exhibit a total lack of dissolution
when tested by standard methodology. In order to enhance the relative
bioavailability of CoQ10, a new formulation and process (BioSolv™,
BioSytes) was used to prepare softgel capsules (Q-GEL™) which
provide 100% dissolution when tested by the USP method. These softgel
cap sules (Q-GEL™) were compared with a compressed tablet, a
powder filled hardgel capsule and a commercially available softgel
capsule in a relative bioavailability study. Twenty-four adult male
subjects were divided into four groups, with the subjects in each
group receiving 120 mg of CoQ10 daily (single dose) of one of four
different formulations for three weeks. Plasma was collected at 0,
1, 2 and 3 weeks for CoQ10 determinations by HPLC. Relative bioavailability
was calculated using the plasma CoQ10 AUC [Dr. Swarwtout's note: AUC
stands for "Area Under the Curve", a method for measuring
relative bioavailability] . The data show that compared with the stan
dard softgel capsule, hardgel cap sule and tablet (all three having
similar relative bioavailability), the Q-GEL softgel formulation is
vastly superior (ca. 300%) (FASEB Journal, 11:A586, 1997).
